Famotidine is a potent an inhibitor of H 2 histamine receptors which main clinically significant pharmacological action is the suppression of stomach juice production. Famotidine reduces both the acidity of the juice and the volume of gastric production, while the production of pepsin remains proportional to the amount of secreted gastric juice.

Famotidine suppresses basal, nocturnal, stimulated by the introduction of pentagastrin, betazole, caffeine, insulin and the physiological reflex of the vagus nerve secretion of the stomach acid.

The duration of inhibition of secretion when using doses of 20 mg and 40 mg is from ten to twelve hours.

Famotidine almost does not affect gastrin levels on an empty stomach or after eating. The drug does not affect stomach emptying, pancreas function, blood flow in the liver and in the portal system.


Famotidine is rapidly absorbed. Oral bioavailability is 40-45%. Bioavailability does not change depending on the contents of the stomach, however, it decreases slightly when taking antacids.

Metabolism during the first passage through the liver has a weak effect on the bioavailability of the formulation.

The maximum plasma level is reached 1-3 hours after the intake. With repeated use, the cumulative effect does not occur. Protein binding is relatively low, 15-20%.

The washing out half-life is 2.3-3.5 hours. In severe renal failure, the half-life of famotidine may exceed 20 hours.

The metabolism of the medication occurs in the liver. The only metabolite found in humans is sulfoxide.
